2-1- تاریخچه غنی سازی با اسیدهای چرب
در اواخر دهه 1960، چندین محقق مشکلاتی را در پرورش موفق لارو ماهیان دریایی و گونه هایی از سخت پوستان که از ناپلی آرتمیا بجز آرتمیای SFB تغذیه می کردند را گزارش دادند (Sorgeloos 1980; Leger et al., 1986). در ابتدا مردم عادی چنین تصور می کردند که میزان بالایی از ترکیبات سمی مانند هیدروکربن های کلرینه و فلزات سنگین عامل ارزش غذایی پائین این آرتمیاها از جمله آرتمیای GSL باشد. در مطالعه مقایسه ای میان 8 نژاد از آرتمیا که در گونه شکارچی کفشک زمستانی Pseudopleuronectes americanus انجام شد، ارزش غذایی متفاوت منابع آرتمیا را ثابت کرد (Klein-MacPhee et al., 1980, 1982). در تحقیق دیگری تفاوت ارزش غذایی در 11 گروه ناپلی آرتمیا SFBبرای میگوی Mysidopsis bahia گزارش گردید (Leger et al., 1985a). یافته های مشابهی توسط Watanabe و همکاران (1978) و Kanazawa و همکاران (1979) در مورد ماهیان دریایی عنوان شد.
2-2- دلایل غنی سازی با اسیدهای چرب
باکیفیت ترین آرتمیا، آرتمیای خلیج سانفرانسیسکو (SFB) یا Artemia franciscana است که بیشترین مقدار EPA را نسبت به گونه های دیگر دارد اما میزان آن به اندازه میزان EPAدر آرتمیای غنی شده با HUFA نمی باشد.SFB از لحاظ DHA کمبود دارد که DHA مهمترین HUFA برای لارو ماهیان دریایی می باشد (Granvil, 2000).
در ترکیب شیمیایی آرتمیا ارومیانا EPA بسیار کم و اسیدچرب DHA در حد صفر می باشد. لذا غنی سازی آن قبل از آنکه به مصرف لارو آبزیان برسد ضروری است (مناف فر، 1380).
نژادهای آرتمیا از لحاظ اندازه و کیفیت تغذیه ای خصوصاً از لحاظ میزان اسیدهای چرب غیراشباع زنجیره بلند (HUFA) با یکدیگر متفاوتند (Granvil, 2000). اکثر نژادهای آرتمیا، نسبت پروتئین به انرژی پائینی دارند که می توان با استفاده از فرایند غنی سازی در ناپلی آرتمیا تغذیه شده با یک منبع غنی (به طور مثال از اسیدهای چرب غیراشباع چند گانه (PUFA)) ارزش غذایی ناپلی را از نظر نسبت پروتئین به انرژی برای موجود هدف بهینه سازی کرد.
ناپلیوس آرتمیای تازه تخم گشایی شده تنها مقادیر کمی از اسیدهای چرب غیر اشباع زنجیره بلند n-3 HUFA را دارد، EPA ممکن است وجود داشته باشد یا کم باشد در حالی که DHA معمولاً در آرتمیا یافت نمی شود (Watanabe et al., 1987). اکثر سویههای آرتمیای مطالعه شده دارای میزان بیشتر از20% لینولئیک اسید (LA, 18:2 n-6) از کل اسیدهای چرب و میزان DHA کمتر از 5%، میزان لینولنیک اسید (Lna, 18:3 n-3) کمتر از 10% و EPA بین 5 و 12% میباشد (Bengtson et al., 1991).
2-3- غنی سازی اسیدهای چرب ضروری در ناپلیوس آرتمیا
در ارزیابی ارزش غذایی غذای مورد استفاده در تغذیه لارو ماهی، چربی ها یک فاکتور کلیدی هستند زیرا چربی ها اساس غشاهای سلولی را تشکیل می دهند، همچنین مسئول حمل ترکیبات مهمی از جمله ویتامین ها و هورمون به درون جریان خون هستند (Koven et al., 1990; Czensy et al., 1999; Copeman et al., 2002). افزایش میزان اسیدهای چرب ناشی از غنی سازی، نشان می دهد که اسیدهای چرب مهمترین نقش را در مقاومت در برابر استرس دارند (Lavens and Sorgeloos, 1996). علاوه بر این، اسیدهای چرب ضروری EFA در کمک به فعالیت سیستم ایمنی و مقاومت به بیماری در چندین گونه از نرمتنان و ماهیان نقش دارد.
ارزش و کیفیت غذا را اجزاء تشکیل دهنده آن تعیین می کند که از میان این اجزاء، نوع و کیفیت اسیدهای چرب، نقش تعیین کننده ای بر کیفیت و قیمت غذا و در نهایت کیفیت لاروهای تولیدی دارند (Watanabe et al., 1982). عامل اصلی اثرگذار در ارزش غذایی آرتمیا، میزان اسید چرب زنجیره بلند غیراشباع (HUFA) ایکوزاپنتانوئیک اسید 20:5n-3 (EPA) می باشد (Leger et al., 1985b, 1987a).
مطالعات زیادی، نیاز به اسیدهای چرب ضروری (EFA) را ثابت کرده اند، و نشان داده اند که نیاز به اسیدهای چرب ضروری به طور قابل ملاحظه ای از گونه ای به گونه دیگر و در داخل گروه ها نیز متفاوت می باشد. قزل آلای رنگین کمان به اسیدهای چرب از خانواده لینولنیک (امگا 3) نیاز دارد (Castell et al., 1972) در حالی که کپور، مارماهی و Chum salmon نه تنها به خانواده اسید لینولنیک بلکه به اسید لینولئیک هم برای رشد خوب نیاز دارند (Watanabe et al., 1982). همچنین میگوی ژاپنی با 1% اسید لینولئیک یا لینولنیک بهترین رشد و بقا را دارد (Kanazawa et al., 1979).
اسیدهای چرب غیراشباع نقش مهمی در فعالیت های زیستی و فیزیولوژیکی آبزیان ایفا می نمایند. مطالعات مربوط به نیاز سخت پوستان دریایی به اسیدهای چرب ضروری، از اواسط دهه 1970 میلادی آغاز شده است (Sorgeloos et al., 1998). بررسی ها نشان داده اند که نداشتن و عدم سنتز سریع اسیدهای چرب غیراشباع زنجیره بلند به ویژه EPA و DHA توسط لارو میگو و ماهیان دریایی، رشد و بازماندگی آن ها را در مراکز تکثیر کاهش می دهد.
میزان اسیدهای چرب ضروری مانند EPA و DHA در غذاهای زنده ای (آرتمیا و روتیفر) که در مراحل اولیه زندگی لارو مورد استفاده قرار می گیرند به طور طبیعی کم است. ناپلی آرتمیا، رایج ترین طعمه پرورشی در آبزی پروری می باشد اما ممکن است نتواند نیاز گونه هدف به اسیدهای چرب ضروری را به طور کامل تامین نماید. آرتمیا از لحاظ EPA فقیر می باشد (Czensy et al., 1999). بنابراین به نظر می رسد که آرتمیا برای تغذیه لاروی، خصوصاً در مقایسه با پاروپایان، زیر حد بهینه باشد (Sargent et al., 1997; Nanton and Castell, 1999; Han et al., 2000; Hanaee et al., 2005).
بنابراین، غنی سازی آن ها با امولسیون های حاوی اسیدهای چرب غیراشباع، ضروری به نظر می رسد (Copeman et al., 2002). Kiron و همکاران در سال 1995 گزارش دادند که اسیدهای چرب 3ω پیش ماده مهمی در سنتز ایکوزانوئیدها هستند که در حقیقت واسطه های مهمی در واکنش های التهابی و تنظیم پاسخ ایمنی بدن می باشند.
غنی سازی ناپلیوس آرتمیا با اسیدهای چرب، نه تنها تفاوت در کیفیت تغذیه ای (غذایی) نژادهای آرتمیا را (مثلاً از لحاظ میزان EPA) به حداقل می رساند بلکه ناپلیوس ها را به غذایی با کیفیت بالا (مثلاً از لحاظ DHA) تبدیل می کند (Leger et al., 1987). غنی سازی غذای زنده از قبیل آرتمیا با چربی های غنی از EFA قبل از تغذیه توسط ماهیان، با توجه به کمبود این مواد در آرتمیا ضروری می باشد (Copeman et al., 2002).
فرآیند غنی سازی ناپلی آرتمیا فرانسیسکانا با روغن های مختلف، به طور قابل توجهی میزان همه اسیدهای چرب را بالا می برد که مهمترین آن ها DHA و EPA هستند، این دو اسید چرب برای رشد و نمو گونه هایی از نرمتنان و ماهیان ضروری هستند (Immanuel et al., 2007).
غنی سازی غذای زنده با اسیدهای چرب ضروری، یک عمل رایج در تفریخگاه های ماهیان دریایی و میگو است (Sorgeloos et al., 1987). روغن ماهی دارای مهمترین اسیدهای چرب (ایکوزاپنتانوئیک اسید و دوکوزاهگزانوئیک اسید) برای میگوی دریایی و لب شور می باشد (Immanuel et al., 2004).
غنی سازی را می توان هنگام انکوباسیون سیست ها با افزودن مواد مغذی به داخل ظروف انکوباسیون اعمال کرد و یا می توان بعد از تفریخ ناپلیوس ها و جداسازی ناپلیوس ها از سایر مواد زاید، در داخل ظروف جداگانه ای، بدین کار مبادرت کرد که در روش دوم میزان غنی سازی HUFA تا ده برابر روش اول هم می رسد (Leger et al., 1987). همزمان با جذب اسیدهای چرب توسط آرتمیا، پروفیل اسیدهای چرب بسته به مدت غنی سازی، تغییر می کند.
امروزه دیگر استفاده از جلبک ها برای غنی سازی آرتمیا توصیه نمی شود چرا که اولاً باید کشت شوند و ثانیاً حاوی مقادیر متغیری از 20:5n-3 و 22:6n-3 هستند. استفاده از مخمر امگا و مخمر نانوایی غنی شده با روغن ماهی شاید مشکل متغیر بودن مقادیر اسیدهای چرب ضروری را برطرف سازد ولی به صورت تجاری در دسترس نیست (Leger et al., 1987). استفاده از غذاهای فرموله و امولسیفای شده که اساساً از روغن های حاوی مقادیر بالایی از HUFA تهیه شده اند امکان بهتری را برای غنی سازی موثر آرتمیا در سطح وسیع فراهم می آورد (Leger et al., 1987).
علائم کمبود اسیدهای چرب ضروری شامل رشد کند، کارایی غذایی پائین، کم خونی و مرگ و میر بالا می باشد (Takeuchi et al.,1979; Roberts and Bullock, 1989; Sargent et al., 1989). چربی های جیره غذایی نقش مهمی را در تغذیه ماهی به منظور تامین اسیدهای چرب ضروری و انرژی ایفا می کنند، همچنین به جذب مواد غذایی محلول در چربی نیز کمک می کنند (Sargent et al., 1999).
